Cancer usually starts out as a pain free lump so chances are that this is not related to cancer. If you are sexually active then this can be related to an STD so you do need to be tested for all STD's. It can also be a pimple as they appear even on the penile area. If this has been there for two weeks and is now starting to hurt you should be seen for a physical examination so that further evaluation can be performed. But this does not sound cancer related it sounds more like infection, pimple, or STD related and it may be that a round of antibiotics are needed.

This could also be related to a boil which can take up to three weeks to come to a head and heal. If you have had this for three weeks, it has started getting bigger and painful this could very well be the cause. Some warm compresses will help this to heal faster but do not try to squeeze or touch this as this can cause the infection to spread. With cancer it would not become painful this soon so it does not sound like cancer, although any unusual moles, bumps, or changes in the skin should have further evaluation.
